The short answer to your question is yes. Of course, without the
specific cards involved unusual circunstances can alter your results, so
I'll use examples.
Circle of Protection: Red

Enchantment
1: The next time a red source of your choice would deal damage to you
this turn, prevent that damage.
and let's say,
Boros Swiftblade

Creature — Human Soldier

Double strike
The fact that the Swiftblade is both red and white does not change the
fact that it is red. Therefore, if you activate the CoP: Red (and choose
the Swiftblade), you will prevent the next damage it would deal you.
Note that you would have to do this twice to stop all the damage it would
do to you during combat, due to it's double strike ability.
Further, note that if the creature was a:
Glory Seeker

your CoP: Red would not work, even if your opponent used a R and a W mana
to cast it, because in the absence of any effect that says otherwise, a
creature's color(s) depends solely on its mana cost, not what you
actually paid for it.
